Selected Properties of Lipase and Protease of Pseudomonas fluorescens 27 Produced in Four Media

ABSTRACT

Lipase and protease were produced simultaneously by Pseudomonas fluorescens 27 in three types of broth and on a dialysis membrane resting on semisolid media. Lipase activity was high from pH 6 to 9, and protease activity was high from pH 6 to 8. Both types of activity were near optimum at 45°C, and each was high at 5°C. Medium in which enzymes were produced had no effect on activation energy. Enzymes produced on dialysis membrane separated into eight visible bands on electrophoresis. Lipolytic activity was associated with each band, but no band was proteolytic. All detectable proteolytic activity was lost during electrophoresis. Lipase was detected by the radial diffusion method (spirit blue agar into which tributyrin was emulsified by sonication) at .1 the concentration detected by the copper soap method.
