Application of Dialysis of Measure Protease Activity in Milk

ABSTRACT

A dialysis technique for measuring proteolysis in milk was investigated. This method consists of adding buffered enzyme substrate mixture to a dialysis bag that is immersed into a test tube containing 10 ml distilled water. After incubation at 37°C for a specific time the bag is discarded. Ten milliliters of 15% sodium carbonate solution is added to contents of the test tube followed by phenol reagent. Color is developed for 15 min before absorbance at 650 nm is measured. This simple procedure had a sensitivity equal to.the Hull method, required less glassware and reagents, and did not form precipitates.

This dialysis test with 30-min incubation also may be used to determine free tyrosine as an index of proteolysis in stored milk. It is as sensitive as the Hull method.
