Chromatographic, Immunochemical, and Milk-Clotting Activity Measurements in Argentine Commercial Liquid Rennets

ABSTRACT

Samples of 25 liquid rennets were obtained from cheese factories in the province of Santa Fé in Argentina. Preliminary analyses were for pH, milk clotting activity, and amylase.

Monospecific antibodies against chymosin, bovine pepsin A, porcine pepsin, and acid proteases from Mucor miehei, Mucor pusillus, and Endothia parasitica were used to identify enzymes in commercial liquid rennets. The immunodiffusion test clearly differentiated the rennets; 18 were mixtures of chymosin and bovine pepsin A, one a mixture of porcine pepsin and Mucor pusillus, and the other samples were derived from Mucor. All Mucor samples were positive for amylase.

Percentages of milk clotting activity due to chymosin and bovine pepsin A were determined by diethylaminoethyl-cellulose chromatography. Chymosin clotting activities were below 20%, indicating that all bovine rennets analyzed were manufactured from adult bovine stomachs.
