Effect of Perchlorate on the Milk-Plasma Radioiodine Ratio of Intact and Thyroidectomized Goats1

ABSTRACT

The purpose of this study was to investigate the effects of graded doses of sodium perchlorate upon the iodide-concentrating mechanism of mammary gland of intact and thyroidectomized goats given twice-daily doses of 125 I over extended periods. Perchlorate was given intravenously at 0.25, 0.5, 1.0, 2.0, 4.0, and 8.0 mg/kg of body weight as a single dose or orally as twice-daily doses for extended periods. The regression equation of the milk/plasma radioiodine ratio on the single intravenous perchlorate dose was Y = 49.5 – 32.0 log X with no difference between intact and thyroidectomized goats. The mammary gland iodide-concentrating mechanism can operate independently of the thyroid. Plasma nonbound iodide was described by the equation Y = 32.3 + 61.2 log X for both classes of goats. When perchlorate was given in twice-daily oral doses for 7 days, the depression of the milk/plasma ratio at the end of this period was greater than after the single intravenous administration of perchlorate.
