-Casein Variants in Buffalo Milk1National Dairy Research Institute, Karnal, Haryana, India
ABSTRACT
The discovery of genetic polymorphism by Aschaffenburg and his associates (1–3) has stimulated much interest in milk protein research (6, 9, 10). Genetically controlled polymorphism in
s-(6), ß-(1, 2, 9) and
-casein (7) from cow milk has been reported. This paper will present evidence for
-casein vari ants in buffalo milk, using a modified starch gel electrophoretic method developed by Ganguli and Majumder (5).
Milk samples were collected from individual Murrah buffaloes in our Institute's herd. Gasein samples were prepared by isoelectric method (5) and solutions made by dissolving in urea (2 g/10 ml milk) in veronal buffer, pH 8.6 according to the procedure of Aschaffenburg (2). Strips of Whatman 3 MM filter paper (10 mm wide) were impregnated with the protein solution, air dried, and stored in a desiccator for use in electrophoresis analysis by the method of El-Negoumy (4). The dried paper strips were cut to convenient size (1.5 by 10 mm) and used directly for analysis.
