ABSTRACT
Milk phospholipids (P-lipids) have been extracted by a new silicic acid column chromatographic (SACC) method (2). This method employs 20% (v/v) formic acid in ethyl ether to elute the P-lipid fraction. Continued investigations on the SACC technique have resulted in several modifications, resulting in more rapid and quantitative recovery.
Raw whole milk samples were obtained from the bulk storage tank in the University Creamery. The extraction methods, manner of handling the samples, lipid phosphorus analysis, and general thin-layer techniques have been previously published (2). One-millimeter chro-matoplates were prepared, using a suspension of 40 g of Silica Gel G (according to Stahl, E. G. Merck, A. G. Darmstadt, Germany) in 80 ml of 0.01 m Na2CO3 (3). The P-lipid spots were identified by spraying the plate with the molybdenum blue reagent of Zinzadze, as reported by Dittmer and Lester (1). All organic material was determined by placing the plate (sprayed with molybdenum blue reagent) in a 110 C oven overnight.
